Monitoring collective cell migration using live-cell imaging

Wound healing is a dynamic process involving many different cell types. One of the most important parts of this process is the collective migration of cells into the wound area. To obtain more insight into collective migration during wound healing, the wound healing assay (also called scratch assay) is mostly used. However many different variations of this assay have been developed throughout the years. Traditionally, wound closure is analyzed at just a few timepoints. Next to this, since the wound closure is often analyzed manually, low sample sizes are used. Recently, research labs realize the importance of analyzing kinetics in wound healing assays. Besides, more automated methods have been developed to increase throughput and reproducibility of the scratch assays. Automated live-cell imaging is one of these methods that have enabled analyzing the kinetics of the wound healing process with high throughput.

Simone Riis-Porsborg, Ph.d., Associate professor at Regenerative Medicine at Aalborg University, Denmark.

  • Presentation title: "Adipose-derived stem cells for the treatment of chronic wounds"
  • Presentation abstract: Chronic wounds affect millions of people, and as the general population advances in age and the prevalence of lifestyle diseases raises, chronic wounds are becoming an increasing health burden. Adipose-derived stem cells (ASCs) are multipotent cells responsible for the reparation and maintenance of tissues. In pre-clinical studies, ASCs have shown great promise as a treatment modality for healing cutaneous wounds. For having an ASC-based advanced therapy medicinal product (ATMP) approved, a measure of potency must be established. Therefore, we develop in vitro assays reflecting the mode of action of ASCs in wound healing, hopefully correlating to the clinical effect of these.

Simone Riis Porsborg is an associate professor within Regenerative Medicine at Aalborg University, Denmark. Simone is paving the way with her research for translating basic stem cell research into clinical use and currently focusing on the use of adipose-derived stem cells for the treatment of diabetic and other chronic wounds.

Hanan Osman-Ponchet, Ph.d., EMBA, Founder & CEO at PKDERM, France

  • Presentation title: "Evaluating wound healing properties of cosmetic ingredients using live-cell imaging with CytoSMART Omni device"
  • Presentation abstract: The wound healing assay is a standard in vitro technique to study cell migration. Although the assay is straightforward and widely applied, it lacks standardization, making it difficult to compare results and reproduce experiments. A standardized wound healing assay developed by PKDERM will be presented. In this assay, cells are cultured in Ibidi® inserts creating reproducible wounds. Live-cell imaging of the wounded area and quantifying cell migration were done using the CytoSMART Omni. Its application in the cosmetic field will be presented through two examples using human epidermal keratinocytes and human dermal fibroblasts treated with different cosmetic ingredients. Overall, the presented wound healing assay provides an efficient and reproducible screening tool with high-throughput experiments.

Dr. Hanan Osman-Ponchet is currently Founder and CEO at PKDERM. The French biotech provides innovative in vitro solutions to evaluate the efficacy and safety of pharmaceutical and cosmetic products. Hanan has a Ph.D. and an executive MBA and a long experience in the pharmaceutical industry. She has authored more than 30 peer-reviewed publications and patents and has given invited oral presentations at different scientific conferences.

Linda Boekestijn, M.Sc., Application Scientist at CytoSMART Technologies, the Netherlands

  • Presentation title: "Development of a high-throughput 3D wound healing model."
  • Presentation abstract: Current therapies for chronic or severe wounds are generally painful, high in cost, and enclose a long and often incomplete recovery. Abundant research is performed to develop new therapies that overcome these limitations. This research is changing from 2D toward 3D models because these models can mimic the native cellular environment. However, 3D models require high technical skills, and they are high-laborious. We developed a high-throughput 3D wound healing model that mimics different phases of the wound healing process in a relatively easy and fast 3D experimental set-up. Here, we used a 3D magnetic bioprinting technique and the CytoSMART Omni.

Linda Boekestijn started as an application scientist at CytoSMART last October. Linda studied Biomedical Engineer at the Technical University of Eindhoven, during the last part of her master's she did her externship at CytoSMART. In this period, she developed a new high-throughput 3D wound healing model.

Inge is a Senior Application Scientist at CytoSMART with a background in life sciences. She obtained her Ph.D. in soft tissue engineering and mechanobiology at the Eindhoven University of Technology. As a Senior Application Scientist, she is involved in the development of new applications for CytoSMART devices and supports customers with their application-related questions.
