Construction of Minicircle Suicide Genes Coding for Ribosome-Inactivating Proteins

Suicide gene therapy is a therapeutic strategy based on the delivery of suicide genes to cancer cells, which then kill themselves by expressing toxic ribosome-inactivating proteins. Plasmid DNA vectors are easy-to-produce, but are known to cause immunological side effects. To counteract this, the minicircle technology can be used. The authors of this article describe a methodology to produce minicircle-DNA at low costs. Following transfection, the cell killing properties were monitored with the CytoSMART Omni and analyzed using the CytoSMART Confluency Module.
